The position of the developing fetus' bladder is very key when identifying a baby's gender using nub theory. A females bladder lies just slightly under the Nub/Developing clitoris whereas a males bladder lies slightly lower in comparison. We use spine line along with the baby's nub and bladder to identify the angle in which we find. This is scientifically proven and when clear can hold up to 99% accuracy in identifying the sex of a baby as young as 12 weeks. Please see below an example of a 2d scan of a baby girl and baby boy around 12 weeks gestation and the scientific findings in comparison.

Here you can see in comparison to the female a male bladder lies lower and therefore creates a much higher angle, which will be above 30 degrees when going to the tip of the nub/ developing penis.
